该文通过双[3-(三甲氧基甲硅烷基)]丙基胺与硬脂酰氯的亲核取代反应,制备双[3-(三甲氧基甲硅烷基)]十八烷基丙基酰胺硅烷化试剂,再通过双齿键合的方式将其修饰到硅胶表面,得到酰胺极性基团嵌入式C
18
双齿键合硅胶色谱固定相(Sil-AM-C
18
)。通过红外光谱和核磁波谱表征,结合元素分析数据,证明双齿酰胺修饰的C
18
硅烷中间体(AM-C
18
)成功合成并键合到裸硅胶上。对Sil-AM-C
18
色谱柱的性能进行了系统评价,结果表明:相较于传统C
18
固定相,Sil-AM-C
18
固定相表现出更高的柱效;由于极性酰胺基团的嵌入,Sil-AM-C
18
固定相具有更好的水相耐受性,在纯水相流动相条件下仍具有稳定的保留和对称的峰形;双齿键合的方式使得Sil-AM-C
18
固定相具有更高的水解稳定性。将Sil-AM-C
18
固定相应用于分离儿茶素类化合物、抗结核药物和维生素C及其衍生物3类亲水性物质,相较于传统C
18
固定相,Sil-AM-C

固定相表现出更优秀的分离能力。
The polarity and water resistance of conventional stationary phases for reversed-phase chromatography can be improved by C
18
stationary phases prepared by embedding an amide polar group in an octadecyl(C
18
) group. Single bonded polar groups embedded in C
18
stationary phases are susceptible to hydrolysis of the silica-oxygen bond,which shortens the service life. In this paper,bis[3-(trimethoxymethylsilyl)]propylamine was prepared by nucleophilic substitution reaction of bis[3-(trimethoxymethylsilyl)]propylamine with stearoyl chloride to produce bis[3-(trimethoxymethylsilyl)]octadecylamido silylating reagent,which was then modified to the surface of silica gel by double-denticle bonding to obtain the amide polar group embedded C
18
double-denticle bonded silica gel chromatographic stationary phase(Sil-AM-C
18
). Characterization by infrared spectroscopy and nuclear magnetic spectroscopy,combined with elemental analysis data,demonstrated that the bidentate amide-modified C
18
silane intermediate(AM-C
18
) had been successfully synthesized and bonded to bare silica gel. The performance of the Sil-AM-C
18
column was systematically evaluated,and the results showed that:compared with the traditional C
18
stationary phase,the Sil-AM-
C
18
stationary phase exhibited higher column efficiency;due to the embedding of polar amide groups,the Sil-AM-C
18
stationary phase had better aqueous phase tolerance,and still possessed a stable retention and symmetric peak shapes under the conditions of pure aqueous mobile phase. The double-tooth bonding of Sil-AM-C
18
stationary phase has higher hydrolytic stability. The Sil-AM-C
18
stationary phase was used for the separation of three types of hydrophilic substances,namely,catechin compounds,anti-tuberculosis drugs,and vitamin C and its derivatives,and showed superior separation ability compared with the traditional C
18
stationary phase.
