采用超高效液相色谱-四极杆-静电场轨道阱高分辨质谱(UHPLC-QE-Orbitrap MS)对天然牛黄和体外培育牛黄共18批样品进行分析,使用Compound Discoverer软件对胆汁酸类成分进行鉴定,结合化学统计学分析筛选不同类型牛黄的差异性成分。根据一级质谱精确质荷比和二级化合物碎片信息,鉴定了30个共有的胆汁酸类成分。聚类分析结果表明体外培育牛黄与产地呈现一致趋势,而天然牛黄的聚类趋势与产地无关。主成分分析载荷图显示胆酸、去氧胆酸等成分对主成分1的贡献较大,而甘氨鹅去氧胆酸、甘氨去氧胆酸、牛磺去氧胆酸等成分对主成分2的贡献
较大。通过建立正交偏最小二乘法判别(OPLS-DA)模型,共筛选出11个胆汁酸类差异成分(VIP

0.05),其中胆酸在两组之间的含量差异最大。除了去氢胆酸和甘氨去氢胆酸,其余9个差异成分在体外培育牛黄中的相对含量均较高。研究结果可为天然牛黄和体外培育牛黄的物质基础、质量控制和资源开发应用提供理论参考。
An ultra-high performance liquid chromatography-Q-Exactive-orbitrap mass spectrometric(UHPLC-QE-Orbitrap MS) method was used to analyse 18 batches of Bovis Calculus(BC) and Bovis Calculus Sativus(BCS) samples. Clustering analysis results indicate that the trend of BCS is consistent with its place of production,while the trend of BC is independent of its source. The load plot of principal component analysis shows that cholic acid and deoxycholic acid contribute significantly to the principal component 1,while glycochenodeoxycholic acid,glycodeoxycholic acid,and taurodeoxycholic acid contribute significantly to the principal component 2. Except dehydrocholic acid and glycodehydrocholic acid,the relative content of the remaining 9 differential components in BCS is higher. This research provide a reference for the material basis,quality control,resource development and application of BC and BCS.
