建立了超高效液相色谱-串联质谱同时测定蜂蜜中胆碱和乙酰胆碱的分析方法。试样采用水-乙腈(体积比2∶3)溶液提取,乙腈-水(体积比8∶2)稀释后以0.22 μm滤膜过滤上机测定。采用ACQUITY UPLC BEH HILIC(100 mm × 2.1 mm,1.7 μm)色谱柱分离,以10 mmol/L乙酸铵(含0.1%乙酸)-乙腈溶液为流动相进行梯度洗脱,采用电喷雾离子源正离子(ESI,+,)模式电离,多反应监测(MRM)扫描,外标法定量。结果表明:在优化条件下,胆碱和乙酰胆碱的检出限(LODs,,S/N ,≥ 3)分别为0.5、0.05 mg/kg,定量下限(LOQs,,S/N ,≥ 10)分别为1.0、0.1 mg/kg;两者分别在0.5~50 ng/mL和0.05~5 ng/mL范围内线性良好,相关系数(,r,2,)大于0.99。空白果葡糖浆(F55)在LOQs、10LOQs、50LOQs 3个浓度水平下的加标回收率为100%~103%,相对标准偏差小于5.0%;在50LOQs浓度水平下,洋槐蜜中的胆碱和乙酰胆碱的加标回收率分别为97.3%和97.8%,相对标准偏差不大于5.1%。该方法前处理简便、快捷,灵敏度高,回收率和重现性良好,适用于蜂蜜中胆碱和乙酰胆碱的测定,已成功应用于70批次蜂蜜和10批次糖浆中两种目标物的测定。
An analytical method was developed for the determination of choline and acetylcholine in honey using ultra performance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS).The samples were firstly extracted with water-acetonitrile solution(2∶3,by volume),then diluted with acetonitrile-water solution(8∶2) and filtered through a 0.22 μm microporous membrance,and finally determined using the UPLC-MS/MS system.The choline and acetylcholine were separated on an ACQUITY UPLC BEH HILIC column(100 mm × 2.1 mm,1.7 μm)by gradient elution,using 10 mmol/L ammonium acetate solution containing 0.1% acetic acid and acetonitrile as mobile phases.The target compounds were analyzed with electrospray ion source in positive electrospray ionization mode under the multi-reaction monitoring(MRM) mode,and quantified by the external standard method.Results showed that,under the optimal conditions,choline and acetylcholine showed good linearities(,r,2,>, 0.99) in the ranges of 0.5-50 ng/mL and 0.05-5 ng/mL,while the limits of detection(LODs,,S/N, = 3) for them were 0.5 mg/kg and 0.05 mg/kg,and the limits of quantification(LOQs,,S/N, = 10) were 1.0 mg/kg and 0.1 mg/kg,respectively.The recoveries for choline and acetylcholine in blank high fructose syrup at three levels of LOQs,10LOQs and 50LOQs ranged from 100% to 103%,with relative standard deviations(RSDs) less than 5.0%,while the recoveries for the analytes in a honey sample at the spiked level of 50LOQs were 97.3% and 97.8%,respectively,with RSDs not more than 5.1%.With the advantages of convenience,sensitivity,good recoveries and repeatability,the method was suitable for the detection of choline and acetylcholine in honey.This method was successfully applied to the analysis on 70 batches of honey and 10 batches of syrup.
