该文通过多种研究方法确定了经典名方清金化痰汤的最佳提取工艺。选取加水倍量、提取次数、提取时间为清金化痰汤制备的关键工艺参数,使用Box-Behnken试验制备了17组清金化痰汤冻干样品。并以物质基准样品的质量作为参照值,计算各组样品与基准样品的基准关联度(SR)。采用层次分析-熵权法(AHP-EWM)即主客观相结合的方法确定栀子苷、芒果苷、黄芩苷、甘草酸、出膏率、指纹图谱相似度的权重系数分别为0.529、0.127、0.083、0.055、0.206、0.000。综合评分后,建立的数学模型具有统计学意义,最终确定清金化痰汤的最佳提取工艺参数为加水6倍,提取30 min,提取1次。重复试验结果显示其综合评分均值为90.865,符合清金化痰汤基准样品质控标准的要求,表明优化后的工艺参数更加稳定可靠,可为后续清金化痰汤颗粒剂的制备奠定基础。此外,响应面结合SR-AHP-EWM的综合试验设计方法为今后其他经典名方的研究提供了思路。
The experiment combined multiple research methods to determine the optimal extraction process for the classical formula Qingjin Huatan Decoction. Based on the concept of quality by design(QbD),critical quality attributes(CQAs),including the water addition ratio,extraction time,and extraction frequency were selected. Seventeen sets of freeze-dried samples of QJHTD were prepared according to the Box-Behnken Design(BBD). Calculated the standard relationship(SR) between each group of samples and the material-based reference sample,using the quality of the material-based sample as the reference value. The analytic hierarchy process(AHP)-entropy weight method(EWM) method,which combined subjective and objective methods,the weight coefficients of geniposide,mangiferin,baicalin,glycyrrhizic acid,yield rate,and fingerprint similarity were determined to be 0.529,0.127,0.083,0.055,0.206,and 0.000,respectively. After comprehensive evaluation,the established mathematical model has statistical significance. The optimal extraction process parameters for QJHTD were determined to be adding 6 times the amount of water,extracting for 30 minutes,and extracting once. The repeated test results showed that the average comprehensive score was 90.865,which meet the requirements of the quality control standard for the material-based reference sample of QJHTD. This indicated that the optimized process parameters were more stable and reliable,laid a foundation for the subsequent preparation of QJHTD granules. In addition,the comprehensive experimental design method combining response surface methodology with SR-AHP-EWM can provide ideas for future research on other classic formulas.
