该文建立了人血清中40余种矿物质元素的快速测定方法。血清样品经含0.01%曲拉通Triton X-100、0.1%硝酸的稀释液直接稀释后,使用电感耦合等离子体质谱仪(ICP-MS)的动能歧视型(KED)碰撞模式进行测定。分别用内标法和外标法进行方法学考察,在选择的仪器工作条件下,大部分待测矿物质元素均获得了良好的线性关系(相关系数

>0.999),检出限为0.000 200~18.6 μg/L。准确度测定结果显示,血清标准品L-1、L-2给出的15种矿物质元素的内标法和外标法检测结果均在认定范围内;对没有给出认定范围的其余元素进行加标回收实验,其加标回收率为89.9%~117%。针对实际血清样品,通过优化稀释浓度和梯度,使用0.1 mL血清样品即可实现含量差异大的多种矿物质元素的测定。所建方法安全、高效、准确,可应用于大样本量人群样本的测定。
A rapid method for the determination of more than 40 mineral elements in human serum was established. Serum samples were pretreated by direct dilution method,the diluent was composed of 0.01% Triton X-100 and 0.1% nitric acid,and then determined by inductively coupled plasma-mass spectrometry(ICP-MS) kinetic energy discrimination(KED) collision mode. The internal standard method and the external standard method were used for methodological investigation. 0.999),and the detection limit was 0.000 200-18.6 μg/L. In the accuracy test,the results of 15 mineral elements of serum standards L-1 and L-2 given by the internal standard method and the external standard method were all within the recognized range. The recovery test was carried out for the remaining elements that did not give the identified range,the recovery was 89.9% -117%. For actual serum samples,by optimizing dilution concentration and gradient,a wide range of mineral elements can be measured using only 0.1 mL of serum sample. This method is safe,efficient and accurate,and can be applied to the determination of large population samples.
