本文基于超表面等离子共振(Meta-surface plasmon resonance
MetaSPR)生物传感器构建了一种新的亲和力检测评价方法,利用高灵敏的MetaSPR芯片,同时构建高精度微流控流路式和高通量微孔板无管路式亲和力检测平台,弥补了现有亲和力检测方法的不足。研究中,制备了具有纳米孔阵列结构的MetaSPR芯片,并通过扫描电子显微镜和酶标仪对其物理结构与光学特性进行表征。通过采用不同折射率的甘油溶液对SPR芯片进行测试,结果显示该芯片对折射率变化具有极高的灵敏度,响应信号与折射率变化之间呈现出良好的相关关系(R
2
=0.995)。利用WeSPR系列检测仪,无需复杂光学元件,对多种分子对进行亲和力检测,成功获得了结合速率常数(
K
a
)、解离速率常数(
K
d
)和解离平衡常数(
K
D
)等动力学参数,且与之前报道的数据一致。结果表明,本研究构建的两种检测平台均能够高灵敏、实时、无标记地监测分子间的结合与解离过程,具有优异的准确性、稳定性与普适性,为生物分子相互作用研究提供了一种高效、灵敏、便携且低成本的技术手段,在生命科学、药物研发与筛选等领域具有广阔的应用前景。
In this study
a new affinity detection and evaluation method is constructed based on the Meta-surface plasmon resonance (MetaSPR) biosensor
which utilizes a highly sensitive MetaSPR chip and simultaneously constructs a high-precision microfluidic flow-through and high-throughput microtiter plate pipeless affinity detection platform
which makes up for the shortcomings of the existing affinity detection methods.methods.In the study
MetaSPR chips with nanopore array structure were prepared and characterized by scanning electron microscop
e and enzyme marker for their physical structure and optical properties.The SPR chip was tested by using glycerol solutions with different refractive indexes
and the results showed that the chip was extremely sensitive to changes in refractive index
and a good correlation was presented between the response signal and the change in refractive index (R2=0.995).The kinetic parameters such as binding rate constant (
K
a
)
dissociation rate constant (
K
d
) and dissociation equilibrium constant (
K
D
) were successfully obtained using the WeSPR series of detector for affinity detection of a variety of molecular pairs without the need for complex optical components and were in agreement with the previously reported data.The results show that both assay platforms constructed in this study are capable of monitoring the intermolecular binding and dissociation processes in a highly sensitive
real-time and label-free manner
with excellent accuracy
stability and universality
providing an efficient
sensitive
portable and low-cost technological means for the study of biomolecular interactions
which is of broad application prospects in the fields of life sciences
