A method for the isolation and purification of (-)-epigallocatechin-3-(3″-O-methyl)gallate(EGCG3″Me) from crude extract sample was firstly developed by high speed countercurrent chromatography(HSCCC).The results showed that HSCCC had a two phase solvent system composing of water-methanol-ethyl acetate-hexane(5∶2∶9∶1,by volume) on a preparative scale.The upper phase was used as the stationary phase,and the lower phase was used as the mobile phase.The revolution speed was 700 r/min,the flow rate was 2.0 mL/min and the detection wavelength was set at 254 nm.Under the optimized conditions,EGCG3″Me can be well isolated.The purity of EGCG3″Me detected by high performance liquid chromatography was more than 90%,and the yield of EGCG3″Me by HSCCC was 76.53%.
