Single-crystal X-ray diffraction(SCXRD) serves as the core technique and gold standard for the structural determination of chiral small-molecule crystals,providing direct,visualizable three-dimensional molecular structures at atomic resolution. Leveraging the anomalous scattering effect of X-rays,SCXRD uniquely and unambiguously determines the absolute configuration of molecules,clearly distinguishing enantiomers(e.g.,
R
/
S
configuration). Using the standard crystal C
12
H
13
Cl
3
O as an example,this study optimized its SCXRD testing conditions by varying five key parameters:data collection strategy,crystal-to-detector distance,data collection temperature,exposure time per frame,and crystal size. The aim was to enhance diffraction data quality and determine the optimal main instrument settings for the structural determination of small-molecule chiral single crystals using the Agilent SuperNova diffractometer.
