基于超高效液相色谱-四极杆-飞行时间质谱(UHPLC-Q-TOF-MS/MS)鉴定术通颗粒49个成分,并利用高效液相色谱-紫外多波长检测器(HPLC-MWD)建立其中12个主要成分的双波长多次切换快速测定方法。色谱柱为Agilent Poroshell 120 EC-C

柱(4.6 mm × 100 mm,2.7 μm),流动相为0.1% 甲酸水-乙腈,梯度洗脱,流速为1.2 mL·min
-1
,柱温为30 ℃,进样体积为5 μL。Dual AJS ESI离子源四极杆-飞行时间质谱在正、负离子模式下分别采集精确分子量及裂解离子。紫外检测采用220 nm及283 nm多次切换(0~2 min、17~21 min、23~25 min,220 nm;2~17 min、21~23 min,283 nm)。通过建立术通颗粒成分数据库,利用精确分子量、碎片离子、对照品比对筛查并鉴定出49个主要的化学成分,并在25 min内实现术通颗粒中12个主要成分的基线分离,定量下限为0.050 0~0.655 0 μg·mL
-1
,加样回收率为98.3%~103%,3批术通颗粒中12个成分的平均含量为0.060~16.597 mg·g
-1
。该方法测定成分多、快速、灵敏、准确,可用于术通颗粒物质基础、质量控制及后续的新药开发研究。
Forty-nine components of Zhutong granules were identified based on ultra high performance liquid chromatography-quadrupole time-of-flight mass spectrometry(UHPLC-Q-TOF-MS/MS). Using high performance liquid chromatograph multi-wavelength detector(HPLC-MWD),a rapid determination method of 12 important chemical constituents in Zhutong granules was established with the aid of multiple dual wavelength switching. The Agilent Poroshell 120 EC-C

chromatographic column(4.6 mm × 100 mm,2.7 μm) was used with 0
.1% formic acid water-acetonitrile as the mobile phase for gradient elution. The flow rate was set at 1.2 mL·min
-1
with column temperature of 30 ℃ and injection volume of 5 μL. Dual AJS ESI ion source quadrupole time-of-flight mass spectrometry was used to collect accurate molecular weight and fragment ion in positive and negative ion modes. Through the establishment of Zhutong granules component database,49 major chemical components were screened and identified by accurate molecular weight matching,fragment ion and reference substance comparison. Dual wavelength switching(0-2 min,17-21 min,23-25 min,220 nm;2-17 min,21-23 min,283 nm) were used to determine 12 main components in Zhutong granules quantitatively within 25 min. Twelve major components reached baseline separation in dual-wavelength switching chromatograms with quantitation limits of 0.050 0-0.655 0 μg·mL
-1
. and recoveries of 98.3%-103%. The average contents of 12 components in 3 batches of Zhutong granules were 0.060-16.597 mg·g
-1
. This method can rapidly determine more components with good sensitivity and accuracy,which is useful for quality control and subsequent new drug development of Zhutong granules.
