建立了液相色谱-串联质谱测定特膳食品中天冬酰胺和谷氨酰胺的方法。样品经直接提取或酶解提取,加入内标后使用polar C

色谱柱(150 mm×4.6 mm,2.6 μm)分离,流动相为乙腈和0.02%甲酸溶液,梯度洗脱,在电喷雾正离子扫描模式下进行定性和定量检测。结果表明:天冬酰胺和谷氨酰胺的线性关系良好,相关系数均达0.999以上,线性范围为0~1 000 ng/mL;液态和固态样本的检出限分别为0.8 mg/100g和4.0 mg/100g,定量下限分别为2.4 mg/100g和12 mg/100g;方法回收率为83.5%~104%,相对标准偏差为0.28%~4.2%(

=6)。该方法简便准确,实用性强,可实现特膳食品中天冬酰胺和谷氨酰胺的精准定量。
A liquid chromatography-tandem mass spectrometry method has bee
n established for the determination of asparagine and glutamine in special dietary foods. Following the process of direct extraction or enzymatic extraction,the internal standard was added and the sample was separated by polar C

chromatographic column(150 mm×4.6 mm,2.6 μm). The mobile phases consisted of acetonitrile and 0.02% formic acid in water. It was detected in a positive ion mode of electric spray for quantitative measurement and qualitative check. The results showed that there was a good linear relationship between asparagine and glutamine with correlation coefficients greater than 0.999. The linear range was 0-1 000 ng/mL. The limits of detection(LODs) were 0.8 mg/100g and 4.0 mg/100g for liquid and solid samples,respectively,and the limits of quantification(LOQs) were 2.4 mg/100g and 12 mg/100g for liquid and solid samples,respectively. The recoveries of the method ranged from 83.5% to 104%,with relative standard deviations ranging from 0.28% to 4.2%(

=6). This method has the advantages of simple pre-treatment,short analysis time,good accuracy,and high practicality. It can be used to precise quantification of asparagine and glutamine in specialty foods.
