Differences Analysis of Chemical Constituents in Sanguisorba Officinalis L. Before and After Carbonized Based on UPLC-Q-Orbitrap-HRMS
This study applied ultra-performance liquid chromatography tandem quadrupole electrostatic field orbitrap high resolution mass spectrometry(UPLC-Q-Orbitrap-HRMS) to analyze the changes of chemical constituents of
Sanguisorba officinalis
L.before and after carbonized,and to screen the different hemostatic components. Waters BEH C

(100 mm×2.1 mm,1.7 μm) column was used to process with 0.1% formic acid water solution and acetonitrile as the mobile phase for gradient elution. The heated electrospray ion source(HESI) was adopted to collect the data in both positive and negative ionization modes. Based on the accurate mass,fragment ions,literature and network database,a total o
f 41 chemical components were identified by matching precise molecular ion masses,MS/MS fragmentation patterns,literature data,and database references. These included 17 phenolic acids,12 flavonoids,9 triterpenoids,and 3 other compounds. Principal component analysis(PCA) and orthogonal partial least squares-discriminant analysis(OPLS-DA) were conducted using SIMCA 14.1 software to screen differential components between

After carbonization,the contents of hemostasis-enhancing components(gallic acid,ziyuglycoside Ⅱ,tormentic acid and ursolic acid) increased significantly,while glucogallin,catechin,brevifolin carboxylic acid,ellagic acid and pomolic acid decreased.
