Determination of Asparagine and Glutamine in Special Dietary Foods by Liquid Chromatography-Tandem Mass Spectrometry
A liquid chromatography-tandem mass spectrometry method has bee
n established for the determination of asparagine and glutamine in special dietary foods. Following the process of direct extraction or enzymatic extraction,the internal standard was added and the sample was separated by polar C

chromatographic column(150 mm×4.6 mm,2.6 μm). The mobile phases consisted of acetonitrile and 0.02% formic acid in water. It was detected in a positive ion mode of electric spray for quantitative measurement and qualitative check. The results showed that there was a good linear relationship between asparagine and glutamine with correlation coefficients greater than 0.999. The linear range was 0-1 000 ng/mL. The limits of detection(LODs) were 0.8 mg/100g and 4.0 mg/100g for liquid and solid samples,respectively,and the limits of quantification(LOQs) were 2.4 mg/100g and 12 mg/100g for liquid and solid samples,respectively. The recoveries of the method ranged from 83.5% to 104%,with relative standard deviations ranging from 0.28% to 4.2%(

=6). This method has the advantages of simple pre-treatment,short analysis time,good accuracy,and high practicality. It can be used to precise quantification of asparagine and glutamine in specialty foods.
