摘要

以萘并呋喃酰肼与8-羟基喹啉-2-甲醛为原料，经缩合反应制备新型荧光探针

。采用高分辨率质谱（HR-MS），核磁共振氢谱（

H NMR）和核磁共振碳谱（

C NMR）对其结构进行表征。在缓冲溶液（HEPES）中，当探针

与Ag

络合后，其蓝色荧光大幅猝灭。基于此，探针

能高灵敏识别Ag

。利用高分辨率质谱（HR-MS）考察了探针

对Ag

的响应机制，并测得

识别Ag

的检出限为5.62×10

-8

mol/L。因探针

对Ag

的识别具有灵敏度高、响应速度快（80 s）和生物兼容性好等优点，故利用荧光校准曲线法和原子吸收光谱法对实际水样中Ag

进行分析，结果证明两种方法测定的结果基本一致，探针可用于实际水样中Ag

的快速定量检测。同时，探针还被成功应用于宫颈癌细胞（HeLa）细胞中对Ag

的共聚焦成像。

Abstract

A novel fluorescent probe

was synthesized by condensation reaction of naphthofuranoylhydrazide and 8-hydroxyquinoline-2-carbaldehyde. The structures were characterized by high resolution mass spectrometry（HR-MS），hydrogen NMR（

H NMR） and carbon NMR（

C NMR），the blue fluorescence of probe

was significantly quenched in the presents of Ag

in HEPES buffer solution.Based on this，the probe

can identify Ag

with high sensitivity. The response mechanism of probe

towards Ag

was investigated by HR-MS. The detection limit of Ag

was 5.62×10

-8

mol/L. Because of its high sensitivity，fast response（80 s） and good biocompatibility，the probe

has been successfully applied to the detection of Ag

in actual water samples by fluorescence standard curve method and atomic absorption spectrometry，the data showed that the two methods had essentially identical results.The probe

was also used to the confocal imaging of Ag

in HeLa cells.
